Znanstvenici u Njemačkoj kažu da su razjasnili uzrok rijetke pojave krvnih ugrušaka povezanih s cjepivima protiv koronavirusa Oxford / AstraZeneca i Johnson&Johnson i vjeruju da bi se cjepiva mogla izmijeniti kako bi se takva reakcija potpuno zaobišla, piše Financial Times, a prenosi Jutarnji.

Rolf Marschalek, profesor na sveučilištu Goethe u Frankfurtu, koji od ožujka vodi istraživanje ovog rijetkog stanja, rekao je da je njegovo istraživanje pokazalo da je problem u adenovirusnim vektorima koji oba cjepiva koriste za isporuku šiljastog (spike) proteina virusa Sars-Cov-2 u tijelo.

Mehanizam isporuke znači da cjepiva šalju tzv. 'spike' protein u staničnu jezgru, a ne u tekućinu citosola koja se nalazi unutar stanice u kojoj virus normalno proizvodi proteine, rekli su Marschalek i drugi znanstvenici u radu objavljenom u srijedu.

Plutajući mutirani proteini

Kad uđu u staničnu jezgru, određeni dijelovi šiljastog proteina se spajaju ili razdvajaju, stvarajući mutirane verzije, koje se ne mogu vezati za staničnu membranu tamo gdje se odvija važna imunizacija. Plutajuće mutirane proteine stanice izlučuju u tijelo, što pokreće krvne ugruške kod otprilike jednog od 100.000 ljudi, prema Marschalekovoj teoriji.

Suprotno tome, cjepiva na bazi mRNA, poput cjepiva koje su razvili BioNTech / Pfizer i Moderna, dostavljaju genetski materijal spike proteina u staničnu tekućinu i on nikada ne ulazi u jezgru.

'Kad se geni virusa nađu u jezgri, mogu stvoriti neke probleme', rekao je Marschalek za Financial Times.

Rijetka reakcija zgrušavanja krvi koja je omela cijepljenje AstraZenecom i J&J-om, zabilježena je kod 309 od 33 milijuna ljudi koji su primili cjepivo AstraZeneca u Velikoj Britaniji, uzrokujući 56 smrtnih slučajeva.

U Europi su najmanje 142 osobe imale krvne ugruške od 16 milijuna primatelja cjepiva.

Kao odgovor na to, upotreba cjepiva AstraZeneca ograničena je ili obustavljena u više od desetak zemalja. J&J je započeo s uvođenjem svog cjepiva u Europi s upozorenjem na pakiranju u travnju nakon kratke odgode zbog zabrinutosti oko ugrušaka.

Postoji 'izravan izlaz'

Ali Marschalek vjeruje da postoji izravan 'izlaz' ako oni koji razvijaju cjepiva izmijene slijed spike proteina kako bi spriječili njegovo razdvajanje.

J&J je već kontaktirao Marschalekov laboratorij kako bi zatražio smjernice jer traže načine kako prilagoditi svoje cjepivo kako bi se spriječilo to spajanje, rekao je.

Spike protein u J&J cjepivu već je bio manje sklon 'spajanju' od spike proteina u cjepivu AstraZeneca, čineći reakciju rjeđom, prema Marschaleku. U SAD-u, osam od 7,4 milijuna primatelja J&J-ovog cjepiva izvijestilo je o rijetkoj reakciji.

'Johnson&Johnson sada pokušava optimizirati svoje cjepivo', rekao je.

'S podacima koje imamo u rukama možemo tvrtkama reći kako da mutiraju te sekvence, kodirajući spike protein na način koji sprječava neželjene reakcije spajanja.'

Marschalek je rekao da njegov laboratorij još nije razgovarao o svojim nalazima s AstraZenecom.

'AstraZeneca nas još nije kontaktirala, pa nismo razgovarali s njima, ali ako to učine možemo im reći što da naprave kako bi razvili bolje cjepivo', rekao je.

J&J kaže: 'Podržavamo kontinuirano istraživanje i analizu ovog rijetkog događaja dok radimo s medicinskim stručnjacima i globalnim zdravstvenim vlastima. Radujemo se pregledu i dijeljenju podataka čim postanu dostupni.'

AstraZeneca nije odmah odgovorila na zahtjev za komentarom.

Neki su znanstvenici upozorili da je Marschalekova teorija jedna od mnogih i da su potrebni daljnji dokazi koji bi potkrijepili njegove tvrdnje.

'Nedostaju još dokazi'

'Nedostaju još dokazi koji bi pokazali kauzalni lanac spajanja spike proteina s trombozom', rekao je Johannes Oldenburg, profesor transfuzijske medicine sa sveučilišta u Bonnu. 'Ovo je još uvijek hipoteza koju treba dokazati eksperimentalnim podacima.'

Marschalek je rekao da je nalaze svog laboratorija predstavio Institutu Paul-Ehrlich njemačke vlade i savjetodavnom tijelu zemlje za cijepljenje i imunizaciju.

Znanstvenici u Njemačkoj kažu da su razjasnili uzrok rijetke pojave krvnih ugrušaka povezanih s cjepivima protiv koronavirusa Oxford / AstraZeneca i Johnson&Johnson i vjeruju da bi se cjepiva mogla izmijeniti kako bi se takva reakcija potpuno zaobišla, piše Financial Times, a prenosi Jutarnji.

Rolf Marschalek, profesor na sveučilištu Goethe u Frankfurtu, koji od ožujka vodi istraživanje ovog rijetkog stanja, rekao je da je njegovo istraživanje pokazalo da je problem u adenovirusnim vektorima koji oba cjepiva koriste za isporuku šiljastog (spike) proteina virusa Sars-Cov-2 u tijelo.

Mehanizam isporuke znači da cjepiva šalju tzv. 'spike' protein u staničnu jezgru, a ne u tekućinu citosola koja se nalazi unutar stanice u kojoj virus normalno proizvodi proteine, rekli su Marschalek i drugi znanstvenici u radu objavljenom u srijedu.

Plutajući mutirani proteini

Kad uđu u staničnu jezgru, određeni dijelovi šiljastog proteina se spajaju ili razdvajaju, stvarajući mutirane verzije, koje se ne mogu vezati za staničnu membranu tamo gdje se odvija važna imunizacija. Plutajuće mutirane proteine stanice izlučuju u tijelo, što pokreće krvne ugruške kod otprilike jednog od 100.000 ljudi, prema Marschalekovoj teoriji.

Suprotno tome, cjepiva na bazi mRNA, poput cjepiva koje su razvili BioNTech / Pfizer i Moderna, dostavljaju genetski materijal spike proteina u staničnu tekućinu i on nikada ne ulazi u jezgru.

'Kad se geni virusa nađu u jezgri, mogu stvoriti neke probleme', rekao je Marschalek za Financial Times.

Rijetka reakcija zgrušavanja krvi koja je omela cijepljenje AstraZenecom i J&J-om, zabilježena je kod 309 od 33 milijuna ljudi koji su primili cjepivo AstraZeneca u Velikoj Britaniji, uzrokujući 56 smrtnih slučajeva.

U Europi su najmanje 142 osobe imale krvne ugruške od 16 milijuna primatelja cjepiva.

Kao odgovor na to, upotreba cjepiva AstraZeneca ograničena je ili obustavljena u više od desetak zemalja. J&J je započeo s uvođenjem svog cjepiva u Europi s upozorenjem na pakiranju u travnju nakon kratke odgode zbog zabrinutosti oko ugrušaka.

Postoji 'izravan izlaz'

Ali Marschalek vjeruje da postoji izravan 'izlaz' ako oni koji razvijaju cjepiva izmijene slijed spike proteina kako bi spriječili njegovo razdvajanje.

J&J je već kontaktirao Marschalekov laboratorij kako bi zatražio smjernice jer traže načine kako prilagoditi svoje cjepivo kako bi se spriječilo to spajanje, rekao je.

Spike protein u J&J cjepivu već je bio manje sklon 'spajanju' od spike proteina u cjepivu AstraZeneca, čineći reakciju rjeđom, prema Marschaleku. U SAD-u, osam od 7,4 milijuna primatelja J&J-ovog cjepiva izvijestilo je o rijetkoj reakciji.

'Johnson&Johnson sada pokušava optimizirati svoje cjepivo', rekao je.

'S podacima koje imamo u rukama možemo tvrtkama reći kako da mutiraju te sekvence, kodirajući spike protein na način koji sprječava neželjene reakcije spajanja.'

Marschalek je rekao da njegov laboratorij još nije razgovarao o svojim nalazima s AstraZenecom.

'AstraZeneca nas još nije kontaktirala, pa nismo razgovarali s njima, ali ako to učine možemo im reći što da naprave kako bi razvili bolje cjepivo', rekao je.

J&J kaže: 'Podržavamo kontinuirano istraživanje i analizu ovog rijetkog događaja dok radimo s medicinskim stručnjacima i globalnim zdravstvenim vlastima. Radujemo se pregledu i dijeljenju podataka čim postanu dostupni.'

AstraZeneca nije odmah odgovorila na zahtjev za komentarom.

Neki su znanstvenici upozorili da je Marschalekova teorija jedna od mnogih i da su potrebni daljnji dokazi koji bi potkrijepili njegove tvrdnje.

'Nedostaju još dokazi'

'Nedostaju još dokazi koji bi pokazali kauzalni lanac spajanja spike proteina s trombozom', rekao je Johannes Oldenburg, profesor transfuzijske medicine sa sveučilišta u Bonnu. 'Ovo je još uvijek hipoteza koju treba dokazati eksperimentalnim podacima.'

Marschalek je rekao da je nalaze svog laboratorija predstavio Institutu Paul-Ehrlich njemačke vlade i savjetodavnom tijelu zemlje za cijepljenje i imunizaciju.

'Iznenadili su se našim nalazima, jer nitko nije razmišljao o problemu spajanja', rekao je.
